Design: triplicate ~0.5 g (wet weight) subsamples were taken from three intervals per sediment core (Deep Hole: 0.0 0.5 cm, 1.0 1.5 cm, 2.0 2.5 cm; Snowgoose Bay: 0.0 0.5 cm, 1.5 2 cm, 3.0 3.5 cm). The samples were first washed once with a saline buffer (10 mM EDTA, 50 mM Tris-HCl, 50 mM Na2HPO47H2O at pH 8.0) to remove inhibitors (Poulain et al., 2015; Zhou et al., 1996), and then DNA was extracted from the samples (and a negative reagent control) with the DNeasy PowerSoil Kit (Qiagen, Hilden, Germany) according to the manufacturers instructions. The quality of the DNA was checked with a NanoDrop 2000 (Thermo Fisher Scientific, Wilmington, DE, USA) and through confirming the amplification of the glnA gene from the extracts (and not from negative control) by PCR and gel electrophoresis. Library preparation and sequencing was completed as a shotgun metagenome by Genome Quebec (Montreal, QC, Canada) with Illumina HiSeq 2500 PE125 in triplicate lanes for each sample.

The Arctic is currently experiencing climate-change related warming, which rapidly affects the environmental constraints on its microbial communities. Yet, our knowledge of the community structure and function of high Arctic freshwater microbes is still lacking, even though they play key roles in nutrient cycling. Here, using high-throughput sequencing, we show that sediment microbial communities in the High Arctic's largest lake by volume, Lake Hazen, are highly diverse and contain novel organisms which genomes we could reconstruct.
